Expertise in Thick Tissue Antibody Penetration

The CLARITY tissue clearing method works on thicker tissue samples compared to what is possible with the current gold standard FFPE method.

Fortunately, at ClearLight Biotechnologies we have expertise in proper antibody titration for thick tissue immunostaining penetration.
We have established a specific approach for immunostaining thicker tissues that results in uniform saturation and optimal antibody binding.

Improving the penetration rate of antibodies into thick tissues increases our ability to visualize cellular and subcellular structure. Our methodologies can achieve high signal-to-noise ratios enabling researchers to see more biology.

Antibody Selection and Feasibility

Sometimes researchers want to explore using an antibody that isn't in our optimized list. Customers choose their biomarkers of interest, and if ClearLight has an optimized antibody then we recommend it. Customers may also request that we use an optimized antibody that is commercially available. If at any point we have to optimize an antibody then we perform an antibody feasibility study. The first step is of course to contact us, and let us know more about your research and biomarkers of interest.

It’s All About Expression

When you choose 3D IHC for your research, it’s important to find the right antibody and the right protocol that will address the questions you have. It all comes down to expression: a given antibody must express in your tissue sample with the signal strength and precision to present meaningful scientific results.

Other common immunostaining techniques, e.g., enzyme-linked immunosorbent assay (ELISA), western blot and chromogenic staining, while “tried and true,” can yield data that is muted and muddied and less precise than desired.

ClearLight Biotechnologies uses multiplexed immunofluorescence staining.
The benefits of multiplexed immunofluorescence staining:

  • Provides definitive antibody expression which is easy to visualize
  • Allows tissues to be stained multiple times with different sets of antibodies yielding nuanced information including relationships between biomarkers and cellular structure
