Immunostaining Buffer

$100.00

This reagent has been optimized for the immunostaining of soft tissues cleared using the CLARITY Tissue Clearing Kit (sold separately). After lipid clearing, the tissue-hydrogel hybrid is both transparent to light and permeable to molecules, enabling immunostaining and imaging. This kit is intended for Research Use Only.

Description

This optimized reagent for immunostaining is used for soft tissues cleared using the CLARITY Tissue Clearing Kit - High Lipid or Low Lipid. CLARITY is a tissue clearing method used to render tissues optically transparent, allowing for non-destructive tissue processing and 3D imaging of both normal and diseased soft tissues. In the CLARITY tissue clearing method, the tissue is first embedded with hydrogel monomers to create a matrix scaffold, then the light scattering lipids are removed. After lipid clearing, the tissue-hydrogel hybrid is both transparent to light and permeable to molecules, enabling immunostaining and imaging using a confocal or light-sheet microscope.

The optimized reagent for immunostaining supports step 4 as shown in the Tissue Processing Workflow diagram below.

Safety Data Sheet for Immunostaining Buffer

Analysis can be performed for select biomarkers, please contact us to find out if your target of interest is one of them. Imaging is currently performed on select fields of view instead of the entire sample.

Submission Guidelines

  • The submitted sample must be pre-fixed in either
    • 4% paraformaldehyde (PFA) or
    • 10% neutral buffered formalin (NBF) for 24 hours.
      • Unfixed samples will not be accepted for lab services.
      • Samples prepared with fixatives other than 4% PFA or 10% NBF will not be accepted and have not been validated with our protocol.
    • The tissue must be noted:
      • perfused or not perfused
    • All submitted samples must meet the noted specifications listed in Tissue Sample Requirement.
      • Samples that do not meet the size, species, or tissue sample type will not be accepted.
    • Samples must be shipped in a sealed container (pick 1)
      • PBS or TBS solution.
    • Shipped samples should be scheduled for delivery to ClearLight Biotechnologies Monday through Wednesday with a provided tracking number.
